NM_001082486.2(ACD):c.851A>G (p.His284Arg) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.H370R variant (also known as c.1109A>G), located in coding exon 10 of the ACD gene, results from an A to G substitution at nucleotide position 1109. The histidine at codon 370 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
