NM_001035.3(RYR2):c.6162G>T (p.Lys2054Asn) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K2054N variant (also known as c.6162G>T), located in coding exon 40 of the RYR2 gene, results from a G to T substitution at nucleotide position 6162. The lysine at codon 2054 is replaced by asparagine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
