Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000722.4(CACNA2D1):c.2327A>G (p.Tyr776Cys), citing Ambry Variant Classification Scheme 2023: The p.Y776C variant (also known as c.2327A>G), located in coding exon 29 of the CACNA2D1 gene, results from an A to G substitution at nucleotide position 2327. The tyrosine at codon 776 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
