NM_000051.4(ATM):c.1844T>G (p.Leu615Arg) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.L615R variant (also known as c.1844T>G), located in coding exon 11 of the ATM gene, results from a T to G substitution at nucleotide position 1844. The leucine at codon 615 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
