Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000051.4(ATM):c.1234T>G (p.Trp412Gly), citing Ambry Variant Classification Scheme 2023: The p.W412G variant (also known as c.1234T>G), located in coding exon 8 of the ATM gene, results from a T to G substitution at nucleotide position 1234. The tryptophan at codon 412 is replaced by glycine, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this alteration remains unclear.
