Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000038.6(APC):c.1259G>A (p.Cys420Tyr), citing Ambry Variant Classification Scheme 2023: The p.C420Y variant (also known as c.1259G>A), located in coding exon 9 of the APC gene, results from a G to A substitution at nucleotide position 1259. The cysteine at codon 420 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, in silico predictors for this gene do not accurately predict pathogenicity. Based on the available evidence, the clinical significance of this alteration remains unclear.
