Uncertain significance for Cystic fibrosis — the classification assigned by Ambry Genetics to NM_000492.4(CFTR):c.1330A>G (p.Ile444Val), citing Ambry Variant Classification Scheme 2023: The p.I444V variant (also known as c.1330A>G), located in coding exon 10 of the CFTR gene, results from an A to G substitution at nucleotide position 1330. The isoleucine at codon 444 is replaced by valine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this alteration remains unclear.
