NM_000400.4(ERCC2):c.1301A>G (p.His434Arg) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.H434R variant (also known as c.1301A>G), located in coding exon 13 of the ERCC2 gene, results from an A to G substitution at nucleotide position 1301. The histidine at codon 434 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
