Uncertain significance for Nephrolithiasis/nephrocalcinosis — the classification assigned by Ambry Genetics to NM_000388.4(CASR):c.2341T>G (p.Cys781Gly), citing Ambry Variant Classification Scheme 2023: The p.C781G variant (also known as c.2341T>G), located in coding exon 6 of the CASR gene, results from a T to G substitution at nucleotide position 2341. The cysteine at codon 781 is replaced by glycine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
