NM_000384.3(APOB):c.6583A>G (p.Lys2195Glu) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K2195E variant (also known as c.6583A>G), located in coding exon 26 of the APOB gene, results from an A to G substitution at nucleotide position 6583. The lysine at codon 2195 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
