Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000321.3(RB1):c.2248G>T (p.Asp750Tyr), citing Ambry Variant Classification Scheme 2023: The p.D750Y variant (also known as c.2248G>T), located in coding exon 22 of the RB1 gene, results from a G to T substitution at nucleotide position 2248. The aspartic acid at codon 750 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
