Pathogenic for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000321.3(RB1):c.1616_1617dup (p.Gly540fs), citing Ambry Variant Classification Scheme 2023: The c.1616_1617dupAA pathogenic mutation, located in coding exon 17 of the RB1 gene, results from a duplication of AA at nucleotide positions 1616 to 1617, causing a translational frameshift with a predicted alternate stop codon (p.G540Kfs*4). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
