Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000302.4(PLOD1):c.1906C>G (p.Gln636Glu), citing Ambry Variant Classification Scheme 2023: The p.Q636E variant (also known as c.1906C>G), located in coding exon 18 of the PLOD1 gene, results from a C to G substitution at nucleotide position 1906. The glutamine at codon 636 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
