Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000268.4(NF2):c.1050A>C (p.Glu350Asp), citing Ambry Variant Classification Scheme 2023: The p.E350D variant (also known as c.1050A>C), located in coding exon 11 of the NF2 gene, results from an A to C substitution at nucleotide position 1050. The glutamic acid at codon 350 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
