Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000264.5(PTCH1):c.3887G>A (p.Gly1296Glu), citing Ambry Variant Classification Scheme 2023: The p.G1296E variant (also known as c.3887G>A), located in coding exon 23 of the PTCH1 gene, results from a G to A substitution at nucleotide position 3887. The glycine at codon 1296 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this alteration remains unclear.
