Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000264.5(PTCH1):c.224C>G (p.Ala75Gly), citing Ambry Variant Classification Scheme 2023: The p.A75G variant (also known as c.224C>G), located in coding exon 2 of the PTCH1 gene, results from a C to G substitution at nucleotide position 224. The alanine at codon 75 is replaced by glycine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
