Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000249.4(MLH1):c.2104-3C>G, citing Ambry Variant Classification Scheme 2023: The c.2104-3C>G intronic variant results from a C to G substitution 3 nucleotides upstream from coding exon 19 in the MLH1 gene. This nucleotide position is highly conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will weaken the native splice acceptor site; however, direct evidence is insufficient at this time (Ambry internal data). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
