NM_000222.3(KIT):c.475G>T (p.Asp159Tyr) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D159Y variant (also known as c.475G>T), located in coding exon 3 of the KIT gene, results from a G to T substitution at nucleotide position 475. The aspartic acid at codon 159 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this alteration remains unclear.
