Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000222.3(KIT):c.1844A>G (p.Asp615Gly), citing Ambry Variant Classification Scheme 2023: The p.D615G variant (also known as c.1844A>G), located in coding exon 12 of the KIT gene, results from an A to G substitution at nucleotide position 1844. The aspartic acid at codon 615 is replaced by glycine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
