NM_000038.6(APC):c.4975G>T (p.Asp1659Tyr) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D1659Y variant (also known as c.4975G>T), located in coding exon 15 of the APC gene, results from a G to T substitution at nucleotide position 4975. The aspartic acid at codon 1659 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, in silico predictors for this gene do not accurately predict pathogenicity. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
