Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000116.5(TAFAZZIN):c.781G>A (p.Glu261Lys), citing Ambry Variant Classification Scheme 2023: The p.E261K variant (also known as c.781G>A), located in coding exon 11 of the TAZ gene, results from a G to A substitution at nucleotide position 781. The glutamic acid at codon 261 is replaced by lysine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
