Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_201596.3(CACNB2):c.472T>C (p.Trp158Arg), citing Ambry Variant Classification Scheme 2023: The p.W104R variant (also known as c.310T>C), located in coding exon 4 of the CACNB2 gene, results from a T to C substitution at nucleotide position 310. The tryptophan at codon 104 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
