Uncertain significance — the classification assigned by Ambry Genetics to NM_199420.4(POLQ):c.641T>A (p.Val214Asp), citing Ambry Variant Classification Scheme 2023: The p.V214D variant (also known as c.641T>A), located in coding exon 5 of the POLQ gene, results from a T to A substitution at nucleotide position 641. The valine at codon 214 is replaced by aspartic acid, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
