Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_198578.4(LRRK2):c.400A>C (p.Ile134Leu), citing Ambry Variant Classification Scheme 2023: The p.I134L variant (also known as c.400A>C), located in coding exon 4 of the LRRK2 gene, results from an A to C substitution at nucleotide position 400. The isoleucine at codon 134 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
