This variant lies in the BRCA2 gene (transcript NM_000059.4) at the canonical splice donor site of the intron immediately after coding-DNA position 8953, duplicating one base. Submitter rationale: The c.8953+2dupT intronic variant, results from a duplication of T two nucleotides after position 8953 in intron 21 of the BRCA2 gene. This nucleotide position is highly conserved in available vertebrate species. In silico splice site analysis predicts that this alteration may weaken the native splice donor site and may result in the creation or strengthening of a novel splice donor site. RNA studies have demonstrated that this variant results in a splice defect; the clinical impact of this abnormal splicing is unknown at this time (Ambry internal data). Based on the available evidence, the clinical significance of this alteration remains unclear.
