Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_177438.3(DICER1):c.640T>A (p.Cys214Ser), citing Ambry Variant Classification Scheme 2023: The p.C214S variant (also known as c.640T>A), located in coding exon 5 of the DICER1 gene, results from a T to A substitution at nucleotide position 640. The cysteine at codon 214 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
