Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_177438.3(DICER1):c.5309A>G (p.Asp1770Gly), citing Ambry Variant Classification Scheme 2023: The p.D1770G variant (also known as c.5309A>G), located in coding exon 23 of the DICER1 gene, results from an A to G substitution at nucleotide position 5309. The aspartic acid at codon 1770 is replaced by glycine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
