Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_177438.3(DICER1):c.4931T>G (p.Ile1644Ser), citing Ambry Variant Classification Scheme 2023: The p.I1644S variant (also known as c.4931T>G), located in coding exon 22 of the DICER1 gene, results from a T to G substitution at nucleotide position 4931. The isoleucine at codon 1644 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
