NM_177438.3(DICER1):c.4545C>G (p.Ser1515Arg) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S1515R variant (also known as c.4545C>G), located in coding exon 22 of the DICER1 gene, results from a C to G substitution at nucleotide position 4545. The serine at codon 1515 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
