Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_177438.3(DICER1):c.3724T>C (p.Tyr1242His), citing Ambry Variant Classification Scheme 2023: The p.Y1242H variant (also known as c.3724T>C), located in coding exon 20 of the DICER1 gene, results from a T to C substitution at nucleotide position 3724. The tyrosine at codon 1242 is replaced by histidine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
