Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_177438.3(DICER1):c.1891A>T (p.Ile631Phe), citing Ambry Variant Classification Scheme 2023: The p.I631F variant (also known as c.1891A>T), located in coding exon 10 of the DICER1 gene, results from an A to T substitution at nucleotide position 1891. The isoleucine at codon 631 is replaced by phenylalanine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
