Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_144997.7(FLCN):c.425T>G (p.Phe142Cys), citing Ambry Variant Classification Scheme 2023: The p.F142C variant (also known as c.425T>G), located in coding exon 3 of the FLCN gene, results from a T to G substitution at nucleotide position 425. The phenylalanine at codon 142 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
