NM_052947.4(ALPK2):c.3170T>G (p.Leu1057Trp) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.L1057W variant (also known as c.3170T>G), located in coding exon 4 of the ALPK2 gene, results from a T to G substitution at nucleotide position 3170. The leucine at codon 1057 is replaced by tryptophan, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
