Uncertain significance — the classification assigned by Ambry Genetics to NM_001386125.1(OBSCN):c.3082G>C (p.Val1028Leu), citing Ambry Variant Classification Scheme 2023: The p.V936L variant (also known as c.2806G>C), located in coding exon 8 of the OBSCN gene, results from a G to C substitution at nucleotide position 2806. The valine at codon 936 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
