Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_032043.3(BRIP1):c.1025G>C (p.Ser342Thr), citing Ambry Variant Classification Scheme 2023: The p.S342T variant (also known as c.1025G>C), located in coding exon 7 of the BRIP1 gene, results from a G to C substitution at nucleotide position 1025. The serine at codon 342 is replaced by threonine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
