Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_021167.5(GATAD1):c.242G>A (p.Gly81Asp), citing Ambry Variant Classification Scheme 2023: The p.G81D variant (also known as c.242G>A), located in coding exon 1 of the GATAD1 gene, results from a G to A substitution at nucleotide position 242. The glycine at codon 81 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
