NM_020774.4(MIB1):c.2731A>G (p.Ile911Val) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.I911V variant (also known as c.2731A>G), located in coding exon 19 of the MIB1 gene, results from an A to G substitution at nucleotide position 2731. The isoleucine at codon 911 is replaced by valine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
