NM_020774.4(MIB1):c.1550A>C (p.His517Pro) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.H517P variant (also known as c.1550A>C), located in coding exon 11 of the MIB1 gene, results from an A to C substitution at nucleotide position 1550. The histidine at codon 517 is replaced by proline, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
