NM_001365276.2(TNXB):c.3226G>T (p.Asp1076Tyr) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D1076Y variant (also known as c.3226G>T), located in coding exon 7 of the TNXB gene, results from a G to T substitution at nucleotide position 3226. The aspartic acid at codon 1076 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
