Uncertain significance — the classification assigned by Ambry Genetics to NM_018975.4(TERF2IP):c.679A>G (p.Asn227Asp), citing Ambry Variant Classification Scheme 2023: The p.N227D variant (also known as c.679A>G), located in coding exon 2 of the TERF2IP gene, results from an A to G substitution at nucleotide position 679. The asparagine at codon 227 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
