Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_017849.4(TMEM127):c.334C>T (p.Leu112Phe), citing Ambry Variant Classification Scheme 2023: The p.L112F variant (also known as c.334C>T), located in coding exon 2 of the TMEM127 gene, results from a C to T substitution at nucleotide position 334. The leucine at codon 112 is replaced by phenylalanine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
