Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_017849.4(TMEM127):c.317G>A (p.Cys106Tyr), citing Ambry Variant Classification Scheme 2023: The p.C106Y variant (also known as c.317G>A), located in coding exon 2 of the TMEM127 gene, results from a G to A substitution at nucleotide position 317. The cysteine at codon 106 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
