This variant lies in the TRPM4 gene (transcript NM_017636.4) at the canonical splice acceptor site of the intron immediately before coding-DNA position 613, where A is replaced by G; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: The c.613-2A>G intronic variant results from an A to G substitution two nucleotides before coding exon 6 in the TRPM4 gene. This nucleotide position is highly conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will weaken the native splice acceptor site and will result in the creation or strengthening of a novel splice acceptor site. Alterations that disrupt the canonical splice site are expected to cause aberrant splicing, resulting in an abnormal protein or a transcript that is subject to nonsense-mediated mRNA decay.
