Uncertain significance — the classification assigned by Ambry Genetics to NM_001166108.2(PALLD):c.2315C>G (p.Ser772Cys), citing Ambry Variant Classification Scheme 2023: The p.S755C variant (also known as c.2264C>G), located in coding exon 12 of the PALLD gene, results from a C to G substitution at nucleotide position 2264. The serine at codon 755 is replaced by cysteine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this alteration remains unclear.
