NM_001166108.2(PALLD):c.1043A>G (p.Asn348Ser) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.N348S variant (also known as c.1043A>G), located in coding exon 2 of the PALLD gene, results from an A to G substitution at nucleotide position 1043. The asparagine at codon 348 is replaced by serine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
