Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_015450.3(POT1):c.926C>T (p.Ser309Leu), citing Ambry Variant Classification Scheme 2023: The p.S309L variant (also known as c.926C>T), located in coding exon 7 of the POT1 gene, results from a C to T substitution at nucleotide position 926. The serine at codon 309 is replaced by leucine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
