Uncertain significance — the classification assigned by Ambry Genetics to NM_001365951.3(KIF1B):c.5281A>C (p.Met1761Leu), citing Ambry Variant Classification Scheme 2023: The p.M1715L variant (also known as c.5143A>C), located in coding exon 44 of the KIF1B gene, results from an A to C substitution at nucleotide position 5143. The methionine at codon 1715 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this alteration remains unclear.
