Uncertain significance — the classification assigned by Ambry Genetics to NM_001365951.3(KIF1B):c.4960A>G (p.Asn1654Asp), citing Ambry Variant Classification Scheme 2023: The p.N1608D variant (also known as c.4822A>G), located in coding exon 43 of the KIF1B gene, results from an A to G substitution at nucleotide position 4822. The asparagine at codon 1608 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
