Uncertain significance — the classification assigned by Ambry Genetics to NM_001365951.3(KIF1B):c.3930T>A (p.Asp1310Glu), citing Ambry Variant Classification Scheme 2023: The p.D1264E variant (also known as c.3792T>A), located in coding exon 34 of the KIF1B gene, results from a T to A substitution at nucleotide position 3792. The aspartic acid at codon 1264 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
