Uncertain significance — the classification assigned by Ambry Genetics to NM_001365951.3(KIF1B):c.3235G>A (p.Glu1079Lys), citing Ambry Variant Classification Scheme 2023: The p.E1033K variant (also known as c.3097G>A), located in coding exon 27 of the KIF1B gene, results from a G to A substitution at nucleotide position 3097. The glutamic acid at codon 1033 is replaced by lysine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
